Generation of anaphors in Chinese
The goal of this thesis is to investigate the computer generation of various kinds of
anaphors in Chinese, including zero, pronominal and nominal anaphors, from the se¬
mantic representation of multisentential text. The work is divided into two steps: the
first is to investigate linguistic behaviour of Chinese anaphora, and the other is to
implement the result of the first part in a Chinese natural language generation system
to see how it works.The first step is in general to construct a set of rules governing the use of all kinds
of anaphors. To achieve this, we performed a sequence of experiments in a stepwise
refined manner. In the experiments, we examined the occurrence of anaphors in humangenerated
text and those generated by algorithms employing the rules, assuming the
same semantic and discourse structures as the text. We started by distinguishing
between the use of zero and other anaphors, termed non-zeroes. Then we performed
experiments to distinguish between pronouns and nominal anaphors within the nonzeroes.
Finally, we refined the previous result to consider different kinds of descriptions
for nominal anaphors. In this research we confine ourselves to descriptive texts. Three
sets of test data consisting of scientific questions and answers and an introduction to
Chinese grammar were selected. The rules we obtained from the experiments make
use of the following conditions: locality between anaphor and antecedent, syntactic
constraints on zero anaphors, discourse segment structures, salience of objects and
animacy of objects. The results show that the anaphors generated by using the rules
we obtained are very close to those in the real texts.To carry out the second step, we built up a Chinese natural language generation system
which is able to generate descriptive texts. The system is divided into a strategic and
a tactical component. The strategic component arranges message contents in response
to the input goal into a well-organised hierarchical discourse structure by using a
text planner. The tactical component takes the hierarchical discourse structure as
input and produces surface sentences with punctuation marks inserted appropriately.
Within the tactical component, the first task consists of linearising in depth-first order
the message units in the discourse structure and mapping them into syntactic-oriented
representations. Referring expressions, the main concern in this thesis, are generated
within the mapping process. A linguistic realisation program is then invoked to convert
the syntactic representation into surface strings in Chinese.After the implementation, we sent some generated texts to a number of native speakers of Chinese and compared human-created results and computer-generated text to
investigate the quality of the generated anaphors. The results of the comparison show
that the rules we obtained are effective in dealing with the generation of anaphors in
Chinese
Knowledge Sharing and Business Matching in Advertising and Public Relations Services Using Semantic Peer Technology
We develop semantic peer network aiming at knowledge sharing and business matching for the domain of advertisement and public relations. We top up a knowledge-based layer upon the peer to peer network to make it knowledge base peer. The knowledge base consists of ontology for the application domain and domain instances. We develop user services for resource sharing and business matching based on the knowledge-based layer. A trust management mechanism is built into the knowledge-based layer for making trustable resource sharing and business match making. Also we develop an RDF-based streaming mechanism for automatically pushing newly matched information to appropriate nodes. We made experiment to test the performance of search for the prototype system. The result shows that the addition of knowledge-based layer upon the peer-to-peer network would not result in the decrease of performance. We also investigate future work after the prototype researc
Prognosis of ductal adenocarcinoma of pancreatic head with overexpression of CD44
SummaryBackgroundThe long-term survival rate of patients with pancreatic ductal adenocarcinoma (PDAC) is very low. Cancer stem cells have been identified in PDAC based on the expression of the surface markers CD24, CD44, CD133, and epithelial specific antigen. The prognosis of PDAC may be related to the presence or absence of tumor cells with cancer stem cell surface markers.MethodsEighty-six PDAC patients (51 male and 35 female patients) who underwent surgical treatment at Chang Gung Memorial Hospital—Lin-Kou Medical Center, Lin-Kou, Taiwan between 1998 and 2007 were included in this study. The patients' ages ranged from 30 years to 84 years. All their surgical specimens showed invasive ductal cancer. Immunohistochemical staining with CD44 antibodies was performed. The differences in clinical data, cell types of tumors, tumor staging, and survival rates between patients with CD44− (Group A; n = 33) and CD44+ (Group B; n = 53) were compared.ResultsClinical data, cell types of tumors, and tumor staging between the two groups showed no significant differences. The 3- and 5-year survival rates were, respectively, 51.5% and 19.8% in patients with CD44− tumor cells and 4.0% and 2.0% in those with CD44+ tumor cells. The differences were statistically significant (p < 0.0001). The median overall survival times of the two groups were also different (36.9 months vs. 12.2 months, p < 0.0001). Multivariate analysis showed that the CD44 as well as lymph node status, and differentiation of tumor cells were prognostic factors for patients with PDAC.ConclusionThe results suggested that CD44 expression in patients with PDAC after surgery was significantly associated with decreased survival, whereas patients with CD44− tumor cells survived significantly longer
The nucleolar protein NIFK promotes cancer progression via CK1α/β-catenin in metastasis and Ki-67-dependent cell proliferation.
Nucleolar protein interacting with the FHA domain of pKi-67 (NIFK) is a Ki-67-interacting protein. However, its precise function in cancer remains largely uninvestigated. Here we show the clinical significance and metastatic mechanism of NIFK in lung cancer. NIFK expression is clinically associated with poor prognosis and metastasis. Furthermore, NIFK enhances Ki-67-dependent proliferation, and promotes migration, invasion in vitro and metastasis in vivo via downregulation of casein kinase 1α (CK1α), a suppressor of pro-metastatic TCF4/β-catenin signaling. Inversely, CK1α is upregulated upon NIFK knockdown. The silencing of CK1α expression in NIFK-silenced cells restores TCF4/β-catenin transcriptional activity, cell migration, and metastasis. Furthermore, RUNX1 is identified as a transcription factor of CSNK1A1 (CK1α) that is negatively regulated by NIFK. Our results demonstrate the prognostic value of NIFK, and suggest that NIFK is required for lung cancer progression via the RUNX1-dependent CK1α repression, which activates TCF4/β-catenin signaling in metastasis and the Ki-67-dependent regulation in cell proliferation

Diversity of the association of serum levels and genetic variants of MHC class I polypeptide-related chain A with liver fibrosis in chronic hepatitis C
Background/Aims Genetic variants of MHC class I polypeptide-related chain A (MICA) at rs2596542 have been associated with hepatocellular carcinoma. The linkage between serum MICA (sMICA) and liver fibrosis in chronic hepatitis C is elusive. Results: Linear regression analysis revealed that sMICA were independently correlated to α-fetoprotein (β: 0.149; 95% confidence interval [CI]: 0.001, 0.003; P = 0.007)and MICA rs2596542 GG genotype (β: 0.209; 95% CI: 0.153, 0.483; P 50 pg/mL provided a positive predictive value of 72 % in predicting advanced liver fibrosis (F3-4) and of 90% in significant fibrosis (> F2) in MICA rs2596542 A allele carriers. Materials and Methods Serum level and single nucleotide polymorphism at rs2596542 of MICA were tested for the association with liver fibrosis in 319 biopsy proven chronic hepatitis C patients. Conclusions: Levels of sMICA were highly correlated to liver disease severity in chronic hepatitis C patients who carried the MICA rs738409 A allele. Patients possessing the genetic predisposition had a higher likelihood of progressed liver fibrosis if they expressed higher sMICA levels
Secretome-Based Identification of ULBP2 as a Novel Serum Marker for Pancreatic Cancer Detection
BACKGROUND: To discover novel markers for improving the efficacy of pancreatic cancer (PC) diagnosis, the secretome of two PC cell lines (BxPC-3 and MIA PaCa-2) was profiled. UL16 binding protein 2 (ULBP2), one of the proteins identified in the PC cell secretome, was selected for evaluation as a biomarker for PC detection because its mRNA level was also found to be significantly elevated in PC tissues. METHODS: ULBP2 expression in PC tissues from 67 patients was studied by immunohistochemistry. ULBP2 serum levels in 154 PC patients and 142 healthy controls were measured by bead-based immunoassay, and the efficacy of serum ULBP2 for PC detection was compared with the widely used serological PC marker carbohydrate antigen 19-9 (CA 19-9). RESULTS: Immunohistochemical analyses revealed an elevated expression of ULPB2 in PC tissues compared with adjacent non-cancerous tissues. Meanwhile, the serum levels of ULBP2 among all PC patients (n = 154) and in early-stage cancer patients were significantly higher than those in healthy controls (p<0.0001). The combination of ULBP2 and CA 19-9 outperformed each marker alone in distinguishing PC patients from healthy individuals. Importantly, an analysis of the area under receiver operating characteristic curves showed that ULBP2 was superior to CA 19-9 in discriminating patients with early-stage PC from healthy controls. CONCLUSIONS: Collectively, our results indicate that ULBP2 may represent a novel and useful serum biomarker for pancreatic cancer primary screening
Connectivity of Default-Mode Network Is Associated with Cerebral Edema in Hepatic Encephalopathy
Cerebral edema, a well-known feature of acute liver disease, can occur in cirrhotic patients regardless of hepatic encephalopathy (HE) and adversely affect prognosis. This study characterized and correlated functional HE abnormalities in the brain to cerebral edema using resting-state functional magnetic resonance imaging (rs-fMRI) and diffusion tensor imaging (DTI). Forty-one cirrhotic patients (16 without HE, 14 minimal HE, 11 overt HE) and 32 healthy controls were assessed. The HE grade in cirrhotic patients was evaluated by the West Haven criteria and neuro-psychological examinations. Functional connectivity correlation coefficient (fc-CC) of the default mode network (DMN) was determined by rs-fMRI, while the corresponding mean diffusivity (MD) was obtained from DTI. Correlations among inter-cortical fc-CC, DTI indices, Cognitive Ability Screening Instrument scores, and laboratory tests were also analyzed. Results showed that gradual reductions of HE-related consciousness levels, from “without HE” or “minimal HE” to “overt HE”, correlated with decreased anterior-posterior fc-CC in DMN [F(4.415), p = 0.000)]. The MD values from regions with anterior-posterior fc-CC differences in DMN revealed significant differences between the overt HE group and other groups. Increased MD in this network was inversely associated with decreased fc-CC in DMN and linearly correlated with poor cognitive performance. In conclusion, cerebral edema can be linked to altered cerebral temporal architecture that modifies both within- and between-network connectivity in HE. Reduced fc-CC in DMN is associated with behavior and consciousness deterioration. Through appropriate targets, rs-fMRI technology may provide relevant supplemental information for monitoring HE and serve as a new biomarker for clinical diagnosis

Clinical Efficacy and Post-Treatment Seromarkers Associated with the Risk of Hepatocellular Carcinoma among Chronic Hepatitis C Patients
This follow-up study enrolled chronic hepatitis C patients to evaluate the treatment efficacy and to identify post-treatment seromarkers associated with risk of hepatocellular carcinoma (HCC) among patients with a sustained virological response (SVR) or nonsustained virological response (NSVR). A total of 4639 patients who received pegylated interferon and ribavirin during 2004–2013 were followed until December 2014. HCC was confirmed through health examinations and data linkage with a national database. A total of 233 HCC cases were reported after 26,163 person-years of follow-up, indicating an incidence of 8.9 per 1000 person-years: 6.9 for SVR and 21.6 for NSVR per 1000 person-years. The associated risk of HCC in patients with SVR was 0.37 (0.22–0.63) for those without cirrhosis and 0.54 (0.31–0.92) for those with cirrhosis compared with their respective counterparts with NSVR. Among patients with SVR, advanced age, male gender, cirrhosis, decreased platelet count, and increased aspartate aminotransferase and α-fetoprotein levels were associated with HCC (p < 0.001). The treatment of chronic hepatitis C patients before they developed cirrhosis showed a higher efficacy than did the treatment of those who had already developed cirrhosis. Patients with SVR may still have a risk of HCC and need to be regularly monitored

Seismic stratigraphy of the central South China Sea basin and implications for neotectonics
Coring/logging data and physical property measurements from International Ocean Discovery Program Expedition 349 are integrated with, and correlated to, reflection seismic data to map seismic sequence boundaries and facies of the central basin and neighboring regions of the South China Sea. First-order sequence boundaries are interpreted, which are Oligocene/Miocene, middle Miocene/late Miocene, Miocene/Pliocene, and Pliocene/Pleistocene boundaries. A characteristic early Pleistocene strong reflector is also identified, which marks the top of extensive carbonate-rich deposition in the southern East and Southwest Subbasins. The fossil spreading ridge and the boundary between the East and Southwest Subbasins acted as major sedimentary barriers, across which seismic facies changes sharply and cannot be easily correlated. The sharp seismic facies change along the Miocene-Pliocene boundary indicates that a dramatic regional tectonostratigraphic event occurred at about 5 Ma, coeval with the onsets of uplift of Taiwan and accelerated subsidence and transgression in the northern margin. The depocenter or the area of the highest sedimentation rate switched from the northern East Subbasin during the Miocene to the Southwest Subbasin and the area close to the fossil ridge in the southern East Subbasin in the Pleistocene. The most active faulting and vertical uplifting now occur in the southern East Subbasin, caused most likely by the active and fastest subduction/obduction in the southern segment of the Manila Trench and the collision between the northeast Palawan and the Luzon arc. Timing of magmatic intrusions and seamounts constrained by seismic stratigraphy in the central basin varies and does not show temporal pulsing in their activities.Keywords: South China Sea, Neotectonism, Core-well-seismic integration, Seismic facies, Seismic stratigraphy, IODP Expedition 34
